Rounding up the latest news from the "black capes"...
'JOKER' GRAPHIC NOVEL [ NEWSARAMA] In October, DC Comics will be releasing 'Joker,' a 128 page hardcover original graphic novel which reunites Brian Azzarello with Lee Bermejo -- Âthe team behind 'Lex Luthor: Man of Steel.' This new tale promises to show a darker, more disturbing side to Gotham City's most notorious villain as he escapes from Arkham Asylum to find that his vast criminal holdings have been split up amongst a number of other notoriously noteworthy villains: The Penguin, The Riddler, and Two Face. With all these villains around -- can Batman be far behind?
Newsarama contacted Brian Azzarello to talk about Joker and what it is about this character that gives evil looking clowns such a bad rap.

I voted for Lex and let me tell you why. First I think the current polling may be influenced by the most recent movie depictions of the Joker and Lex. In that regard I believe Heath Ledger's performance has redefined how people look at the Joker forever, where Kevin Spacey's performance, while IMHO was great, was just a rehash of every other interpretation of Lex Luthor over the past 30 years.
I posit that both characters are criminally insane psychopaths, but that Lex has a better grasp on reality and the ability to keep on task a lot better than the Joker. Granted the Joker may have a plan his erratic nature will cause him to go off plan rather quickly making him unpredictable. While in some cases this unpredictability may be an advantage but over the long term he will go for the laugh first, where Lex will always go for the win.
I think the Joker might beat Lex in a short term fight, but long term, with planning and resources coming into play, Lex will always win (except against Superman of course....but his day will come, his day will come. MARK MY WORDS SUPERMAN! You are facing the greatest criminal mind of our lifetimes, you will eventually fall!).

Personally I think the Joker would win. And I think Jack Nicholson's version would be a better one due to his affinity for poisoning beauty products. Lex Luthor would end up buying said beauty products (mostly because he's a narcissist when you get down to it) and then Lex would mix the products that have the poison in them and then atleast he'd die with a smile on his face.

Joker, without the shadow of a doubt. Superman can give thanks that he has not to face Joker, because indeed as in the last movie is mentioned, all his strength, all his power is just futile against the Joker.
But, forget the Dark Knight. Forget Ledger's brilliant performance.
As read in the brilliant TV Tropes page (if you don't know about it, go check it, it's worth the read) it's just a wonder why the Joker has not been killed. Not by Batman or by any other superhero, but by villains that have worked with him and are pretty powerful and at the same time can be the Noble Demon type.
But the answer is dramatically simple: They are just plain afraid of him. Well, terrified would actually fit better. And why is that? Simple again: When the supervillains gather and want to scare each other in a dark night, they tell each other Joker stories...

I preferred the Mark Hammil version of the Joker. You should try renting Batman Beyond: Return of the Joker (Directors Cut). Or watch any episode with the Joker from Batman the Animated Series to JLU.
